こんにちは。東京理科大学のiGEMチーム、”坊ちゃんLab.”(大会登録名”Botchan Lab Tokyo")です。
私たちは東京理科大学の課外活動団体として創立して3年目、理工学部応用生物科学科の先生方のご指導の下、iGEM大会での受賞を目標に、日々活動しております。

Hi! We’re Botchan Lab Tokyo, iGEM team of Tokyo University of Science. We established this team three years ago, and under the leadership of professors, we’re working hard to get the awards at the iGEM jamboree.


▼iGEM出場は一生ものの経験

iGEMとは、the International Genetically Engineered Machine competitionという合成生物学の学部生主体の世界大会です。

遺伝子組換え技術などを用いて新たな機能を持つ生命システムを作り、その有用性やアイデアの独自性を競います。

マサチューセッツ工科大学(MIT)の授業の一環として始まったiGEMは、昨年の大会で世界各国から318チームが参加する大きな大会になりました。

昨年、iGEM2018への参加に向けて、生物学に限らない幅広い専門知識はもちろん、プレゼンテーション力・コミュニケーション力・世界中のハイレベルな仲間とのネットワークなど、一生ものになるであろうものをたくさん得られました。

特に自分たちと同世代の仲間の発表は、非常に大きな刺激を受けるものばかりでした!

一昨年は銅賞昨年は銀賞を受賞することができ、その経験を生かして本年もiGEM大会に出場し3年目の受賞および、世界を舞台に当団体のさらなる成長、拡大をしたい!と思っています。

The International Genetically Engineered Machine competition(iGEM) is an international team competition made up of predominantly undergraduate students interested in the field of synthetic biology.

By using a technology of genetic engineering, we build new biological devices and systems, and compete for various awards at the iGEM Jamboree.

iGEM began in January of 2003 with a month-long course at MIT, and summer competition began in 2004 with 5 teams. Last year, 340 teams from 42 countries participated in the iGEM Jamboree.

Botchan Lab. participated in iGEM Jamboree since2017, and we inspired by teams whose members are around the same age. We acquired broader knowledge and understanding of synthetic biology to be sure, but also learned approach from different points of view and perspectives by using different academic subjects.


▼テーマは「変異に強い大腸菌」

本年は「変異に強い大腸菌の作製」に挑戦しています。

遺伝情報を担うDNAを安定に保持し、後代に伝えていくことは、生物が種を存続させるためにとても大切です。しかし、DNAを傷つける原因は、放射線、食物中の発がん物質、環境中の化学物質など、身近なところに沢山あるため、DNAは常に損傷の危険にさらされています...。

そこで注目した細菌が、Deinococcus radioduransです。

この菌は通常地上では存在しないほどの強度の放射線にも耐えることができます!他にも紫外線、乾燥、高温、低温など様々な条件にも強い菌です。

なぜ、そんな厳しい条件に耐えられるのか?

それは、この菌はDNAがズタズタに傷つけられても、治す能力がとても高い為です。

そこで今回、この高いDNA修復能力に関係する遺伝子を大腸菌に導入し、「変異に強い大腸菌」を作製しようと考えています。


This year, we’re trying to create “a mutant-resistant E.coli”. 

It is very important for the survival of the species to keep DNA stable and transmit it to the progeny accurately. However, DNA is always at risk of damage because there are many factors of damaging DNA like radiation, carcinogens, chemicals in the environment, ROS and so on. Therefore, cells have the mechanism for repairing their DNA. 

We spotlight “Deinococcus radiodurans” which is known as one of the most radiation-resistant organisms and it can survive cold, dehydration, vacuum, and acidic condition. D. radiodurans accomplishes its resistance to radiation by having multiple copies of its genome and rapid DNA repair mechanisms. 

So, we’re trying to introduce genes related to the high DNA repair ability of D. radiodurans into E.coli, and create E.coli with high DNA repair ability.


▼変異に強い大腸菌は何に応用できるのか 〜宇宙での利用・長鎖DNAの合成〜

1. 宇宙コロニーの新たな食料源

宇宙コロニーとは、宇宙空間に作られた人工の居住地のことです。地球全体での人口の増加・資源の枯渇に対する解決策の1つとして提唱されています。

ですが、実現には様々な課題があり、その1つに自給自足に必要な食料の生産方法を考える必要があります。

そこで、昨年までテーマ(詳細は下に記載しています)にしていた「アミノ酸を合成できる大腸菌」と今年のテーマである「変異に強い大腸菌」の性質を組み合わせることを考えました。

宇宙で生存できる「変異に強い大腸菌」が、アミノ酸を合成できれば、タンパク質の供給源になりえます。

また、宇宙という閉鎖的空間での技術の確立を目指すことは、同時に、地上では災害などでライフラインが絶たれた被災地でも利用可能である為、社会的課題にも貢献します。

A space colony is an artificial residence created in space and is proposed as one of the solutions for population growth and resource depletion on a global scale. However, in order to put it into practice, there are various problems. One of them is the way to produce the food needed for self-sufficiency.

Therefore, we considered combining the previous project: E. coli that synthesizes amino acids, with this year project: E. coli which have resistant to mutation. If E. coli which have resistant to mutations can survive in space and synthesize amino acids, it will be a source of protein.

At the same time, aiming to establish the technology in a closed space like space also contributes to solve social challenges as it can be used in disaster areas where lifelines are stopped by the disasters..


2. 長鎖DNAの合成

これは、生物を使った技術・実験をもっとやりやすくするのに役立つのではないかと考えています。

少し、専門的な話をします。

医薬、創薬の分野だけでなく、農業、工業、環境のあらゆる分野で、生物に新たな遺伝情報を組み込むことで、物質・エネルギー生産系の効率化や、環境にやさしい化合物、新規な化合物を生産しようと新たなバイオプロセスに関心が高まっています。

これを実現しようとすると、そのバイオプロセスに必要なすべての遺伝子を導入しなければなりません。

しかし、操作回数が多くなってしまったり、多種類の選択マーカーが必要になったり、致死性の中間代謝物質が蓄積したり…と、問題点がたくさんあります。

そこで導入する遺伝子群を全て1つのDNA分子中に連結する、すなわち長鎖DNAを合成することができれば、これらの問題点が大幅に改善されるのではないかと考えています。

If we could success above, such an E.coli can help to synthesize long-chain DNA in their cells. In the various fields, there’s interest in new bioprocesses to produce material or energy production systems that is high efficient, environmentally-friendly compounds and new compounds. If you do so, you have to introduce all the genes necessary for the bioprocess. However, there are many problems, such as the increase in the number of operations, the need for various types of selection markers, and the accumulation of lethal intermediate metabolites. Then, if you can synthesize long-chain DNA which links all of the genes that plan to be introduced, it may contribute to solve the problems mentioned before.


▼昨年のテーマ 〜バクテリアルサプリメント〜

昨年までのテーマですと、”バクテリアルサプリメント” でした。

窒素固定能を持たせた大腸菌により、空気中の窒素をアミノ酸に変換させて世界の食糧危機を解決することができるのではないかと考えました。
まず、大腸菌にニトロゲナーゼを発現する遺伝子を導入し、それを発現させることで空気中の窒素からアンモニアを合成しました。続いてアミノ酸脱水素酵素を発現する遺伝子を導入し、その活性によりアンモニアをアミノ酸に変えることを目指しました。

そして将来的にこれをサプリメントの形にしてお腹の中に入れれば、世界の食糧危機、特にタンパク質の面は解決できると考えました。



この研究内容で、iGEM 2017, iGEM 2018に参加し、それぞれ銅賞、銀賞を受賞しました。

Through this project, we got bronze and silver medal in iGEM 2017 and iGEM 2018.

▼最後に

iGEM大会は本当に刺激的な仲間が集まり、非常に魅力的な反面、多くの費用がかかってしまうのも事実です。参加にあたり、チーム登録費として約50万円がかかり、さらに他試薬や菌体の購入が必要です。また、iGEM大会は毎年アメリカのボストンで開催されるため、渡航費や宿泊費もかかります。

多くの費用や時間はかかりますが、それでも有意義な経験が詰まったiGEM大会に、今年も出場し、出場するからには本気で金賞を狙いたいと思っています。

大会への参加を申し込んだため、本プロジェクトはAll-in方式で実施します。目標金額に満たない場合も、計画を実行し、リターンをお届けします。

In order to participate in iGEM Jamboree, we need to prepare for registration fee:$5000 USD. In addition, we need to prepare for the experiment fee. We are undergraduate students and don’t have much opportunity to raise funds. We’d like to challenge ourselves to try to get a gold medal and your help let us challenge this awesome competition.

    新入生も交えて、iGEMモデリング講座を実施しました!モデリングとは、コンピュータを使って実験値の予測を行ったり、実験が正しかったかを確かめたりします。今回は講師にiGEM 2017でモデリングを担当していた当サークルOBさんをお招きしました。大変お勉強になりました!実験の方も頑張りたいと思います!
